I'm going canoeing/camping in the Florida Everglades and I will have no cell signal, but I want to use GPS. There is a program called EarthNC for $9.99 in the market that I can download and cache navigation charts.

What's the best way (ROM, kernel, SetCPU, etc.) to turn off all data and just have GPS active when necessary so that my battery lasts as long as possible?

I know I can toggle the GPS off and on, but should I turn the phone off when not using it or govern my standby frequency to 125MHz and not waste battery by booting the phone every time I need it.

The most important is to switch the phone to "Airplane mode" when not using it. Searching for network (when no network is reachable) can drain the battery in few hours (I know it well, work on powerplants without signal and phone lasts not one day when not in airplane mode).
You can leave GPS enabled, it will be turned on just when needed by some program (but check that no such program is running in background using GPS all the time).
Switching off depends on how many times will you use the phone. If you need it every 6 hours, you can switch it off and boot. If you need it every hour, it will be better to leave the phone on (in airplane mode and gps off).

My personal favorite for this situation is Orux Maps. There are several different maps to chose from and now you can download and cache the maps with your phone. And it's free

But personally, sometimes I have bad dreams that consist of me getting into water with my droid and ruining it forever . . . . no joke. It hasn't happened, but it has been a re-occuring nightmare, lol.

If you put your phone in a ziploc baggie, you can still use the touchscreen, AND all the buttons. Just sayin'.
