Nope, the backup ( CWM or Titanium whatever you used ) is stored on your SD-Card, so as long as you don't partition or format your SD-Card, your backup is safe !!!

........wellllllll, however, it it ALWAYS a good idea to copy a successful backup from the SD-Card to your computer, just in case ( SD_Card's can go bad too, accidental deletes, etc..... )........ I have like 4 - 5 different backups saved on my computer. CWM and Titanium backup........ Of course, I am a backup-freak anyway......

When you make a cwm backup, you can rename them WITHOUT SPACES. i do one a week and then more if i make major changes. This way I'm always up to date incase something happens. If I make tweeks or changes is a great idea before and after, if mess up can revert back and if works then have it if decide to do something else to mess things up. Titanium, I only backup apps, never use to backup system, CWM does that the best.
I also backup the entire sd card and save to pc and then delete the old one every once in a while for same reason that it might go bad or mess up.
