When you plug your phone in via USB, are you asked to "Turn on USB storage"? If not, go to Menu>Settings>Applications>Development>and check USB Debugging. A screen should then pop up asking if you want to turn on USB storage.
Also check in the notification bar. If you see the USB icon while it is plugged in, drag down the bar and tap on USB connected. That should also give you the option to turn on the USB storage.
What phone, and what version of Android are you using?
