I have a nexus S 2.3 unlocked so my question is will i re-lock it if i update my phone??

No, the Nexus line is always unlocked. Updating the firmware does not alter carrier locks on other phones either. For instance, the HTC Desire or Samsung Galaxy, once unlocked they remain unlocked. So feel free to update the phone whenever you want.
