No doubt whatsoever.
Heath Ledger in The Dark Knight.
The most intense amazing performance I have ever witnessed. I never knew he was capable of that. He is sadly missed.

Jack Nicholson - "One Flew Over the Cuckoo's Nest"
Anthony Hopkins - "Silence of the Lambs"

RDJ as Chaplin was pretty spiffing.

Honestly though its too hard to say. Heath was good as Joker. he was also great as Skip in Lords of Dogtown.

Al Pacino - Carlito's way
Morgan Freeman / Tim Robbins - Shawshank Redemption
Kevin Spacey - Life of David Gale / American Beauty
Adam Sandler - Reign Over me
Roberto Benigni - Life is Beautiful
Yolande Moreau - Seraphine
Alan Rickman - Robin Hood, Prince of Thieves

If we threw in TV...

James Gandolfini - Sopranos
John Hamm - Madmen
Michael C Hall - Dexter
Hugh Laurie - House
Edward James Olmos / Mary McDonnell - Battlestar galactica

I would have said Chaplin, but Less Than Zero is a close second, especially considering his life followed a similar path.

Just to name one, Naomi Watts in Mulholland Dr. The scene where she is an actress auditioning for a part and she just transforms into another character before your eyes and you just believe it. Should be a clinic for acting students.

I never thought his performance as the Joker was nearly as impressive as many others seemed to think it was. This isn't to say it wasn't a good performance. He played the role well enough, made you understand and believe the character.

I've heard that Mr. Ledger found playing this role very traumatic, along with speculation that the trauma of playing this role contributed to his subsequent suicide. I don't know how much credence to give any of this. Surely, he's not the first or the best actor to play a monstrous villain, and others who have done so seem to get along just fine afterward. Which leads us to…

Anthony Hopkins - "Silence of the Lambs"
On seeing this thread, I came into it just to mention this performance. Hannibal Lecter wasn't even supposed to be as major a character as he turned out to be.

See an earlier move, titled, “Manhunter”, in which Hannibal Lektor (different spelling than was used in the books or other movies) was played by a minor actor, as a minor character in that film.

When “The Silence of the Lambs” was made, as a sequel to “Manhunter”, Hopkins played the role of Hannibal Lecter, and he played it with such brilliance that the character became much bigger than he was intended to be, such that subsequent stories in that series were specifically written more about Lecter as the main character.

“Manhunter” was even later remade, as The Red Dragon, with Hokins as Lecter; prompting movie critics to complain about how minor a role Hopkins had in that film.

Anthony Hopkins is certainly a very good, top-tier actor. Every part I've ever seen him in, he's played brilliantly. But Hannibal Lecter seems to be by far his greatest role, his greatest performance.

I feel compelled to mention a disappointing performance.

Johnny Depp as Sweeny Tood: The Demon Barber of Fleet Street.

Johnny Depp's not a bad actor, but I think this part was just beyond his abilities. He just didn't seem to have it in him to express the deep, dark, seething emotional rage that this role demanded.

Compare to George Hearn's performance of this role in the 1982 version.
