Are you going to fully charge your Transformer Prime before you use it? Fully as in don't turn it on at all, leave it on the charger for a few hours after it says it's full. I want to but I don't know if I'll be able to hold out after all the waiting just to get it. I don't think it is even necessary, but I want to make sure I am getting the most out of my tablet.

I know I will. I want to be able to spend a good deal of time playing with it, so a full charge it is!

I don't think charging the battery to full before playing will cause any issues. Anyways, most batteries will have a few days and charges before you see an actual battery difference.
So, play with the phone when you get it.
My other suggestion would be to charge the tablet to full, 100%. Drain the battery completely until the tablet turns off 0%. Then, plug the phone into the charger and charge completely and do not unplug the tablet until it's 100%. I would do this a few times after you get it. And since you won't be able to reset the battery stats, this would be the closest thing to recalibrating the battery

When I got the original transformer it had no battery charge at all and I had to charge it to do the update. Not sure if it will be different with the prime.

I like this suggestion and it is what I will be doing when I get my Prime. I've seen this same recommendation posted elsewhere so perhaps it's worthwhile.
I did it with my SGS when I got it. It hasn't had any battery problems and I've had it for over a year now. Can't hurt,....and is probably a wise move.

Everything I've ever read about the Li-ION batteries suggests that complete discharges of the battery actually shortens its life. Also it takes a few days of charging/recharging before you typically see optimal battery performance. I do understand that sometimes battery calibration can get out of whack so the full discharge/charge method is a way to get the battery to calibrate properly. I wouldn't do it unless I had an issue though.
I will use my Prime (if it has any charge at all) when I get it, and put it on the charger either at the end of the day or when I've run it down, whichever comes first.

this is true, but phones and tablets shut off long (relatively) before the battery ever reached 0%. as it is, using a phone or tablet until it reaches 0 will just calibrate the phone or tablets measurement of the battery level--making it more accurate.

Like to say that I'll charge it for eight hours, but I don't know. Probably be too excited. Maybe it will get its full charge when I finally go to bed
