Powering off N5 from lock screen.

Is it just me or when on the lock screen, before I enter my pattern to unlock, I hold down the sleep button and turn the phone off!

This kind of makes Android device manager useless, as if anyone steals phone, they can just turn it off straight away, so that it cannot be traced.

Looking at the Android Device Manager help page, it is not clear what it will do (or not do) when a powered off device is powered back on. I would hope it would act similarly to Lookout (which I have been using since long before there was an ADM). Lookout can't do anything with a powered off device either, but if you make a request to locate a device, it keeps checking. If the device is powered back on and connects to the network, it will locate the device and send you an email.

However, that said, all of these security tactics are fragile. Better than nothing, especially when they are free, but you probably shouldn't depend on them. I have used Lookout's scream feature to help my wife who could lose an 18 wheeler if given the opportunity. ;)

Even if your device is powered off, if you choose to lock it in ADM, when the device is powered on, it will lock with the code you entered in ADM.

It does not happen instantly, but within about 20 seconds of being powered on, even if you have chosen to not use a lock screen at all.

If you use a lock screen, obviously the lock screen would be there first, then the ADM lock screen would take precedence over that. In either case, unless someone knows your normal lock screen unlock pin/gesture/whatever your phone is protected.
