Sorry if it's a dumb question. I'm a novice, but looking to install CyanogenMod 10.1 and the first step says "Make sure you're running ICS bootloaders!".

How can I tell if I'm running one?

The bootloader is the code that tells the operating system to load.

    Two questions...

    1. Are you on ICS, Android 4.0.?, now?

    2. If so, have you flashed any other custom ROM's since being on ICS?

Yes, I'm on 4.1.2 right now. It's the Official Samsung leaked version of Jellybean.
The above was the only time I've flashed a ROM and I did it using a very simple to follow guide I found on here.

So now I want to get to 4.2.1.

In that case I'm guessing that you now have a JB boot loader. You will need to flash an ICS stock ROM via Odin I think to get the ICS boot loader.

Be advised that the CM 10's are all, "Nightly's", to date and are therefore experimental, (work-in-progress),... as opposed to the Stable and Release Candidates.

    CyanogenMod10.1:GT-I9100:Nightly:Known issues
Audio: not all features implemented yet
    USB: slow ums transfer speed
    Camera: recording effects broken
FM Radio: unsupported, will probably never be
TV Out: unsupported, will never work (proprietary, undocumented)
NFC: no support on GT-I9100P is missing
Calling emergency numbers on the following carriers will NOT work: Telia Sweden, Vodafone NL, and KPN Netherlands

    Source, here.

Ironass is indeed correct you'll need the ICS bootloader and you'll need to flash it it in ODIN.

I HAVE NO IDEA WHATSOEVER IF YOU CAN JUST FLASH THIS ON TOP OF YOUR CURRENT OFFICIAL SAMSUNG JB ROM, I WOULD ERR ON THE SIDE OF CAUTION AND GET OTHER ADVICE

Here is the boot loader don't worry that it say "new" it means new for ICS not as in brand new :
