Rooted with autonooter. I lose about 20% of the battery life overnight on stand-by. Is this normal? Seems a little extreme.

Does your WIFI sleep when you turn off the Nook?

If not, there may be an account (Facebook, Twitter, Weather) that its pulling for data often, which will use up battery.

If it does sleep, then there may be an app hanging up causing the Nook to not slep properly, or just using resources in general.

You can always download Spare Parts and see what is using the battery/CPU the most.

I assume that the wi-fi does sleep when the nook sleeps because when I take wake up the nook by pressing "n" I can see that the wi-fi indicator isn't there and I have to wait a second or two for it to show up.

I'll give the spare parts thing a try.

Something is definitely accessing wifi for data. My nook uses next to nothing overnight. I don't run any widgets or background services.
