It isn't entirely clear what you mean by this as Android doesn't really have a default music player.
I have a PlayerPro (my preferred music player) widget on my home screen and I access all my music that way. If you want to be able to click on an audio file in a file manager (although it's not clear to me why you would want to), just do so and you will be asked which app you want to use to open the file with. If you select one and tap on Just Once, you will be asked this question every time you try to open that type of file. If you tap on Always, that app will be the default handler for that file in future.
