I just got started a couple of days ago. It's awesome.
At the moment I use it to bump up the display timeout for certain games I play that require thoughtful pauses (e.g. Daily Sodoku).
I *really* wanted to fix the hideously broken Android "Silent Mode" so that it was actually silent - i.e. clamp down Alarm and Media Volume also, and then restore them to their previous values when Silent Mode was ended. But because the %SILENT variable isn't "dynamic" there's no visible event to handle when Silent Mode is set. I suppose I could set a timer event and poll for it to change state periodically (which is how Tasker internally handles App contexts - it polls every 500ms). I may give that a try even though I hate polling due to resource impact.
I have an action that does the right thing currently, and can associate it with the camera button event, but unfortunately Tasker can't handle hardware events when the screen is locked. It works fine when the screen is unlocked, but it's a hassle to unlock the device just to turn on Silent Mode, plus I want the camera button to actually take pictures when the phone is unlocked

I also plan to set up some location profiles to automatically silence my phone when I go to church, and maybe turn on WiFi when I approach my neighborhood and turn it off when I leave.
I also want to see if I can figure out a way to nudge Google Latitude to update my location periodically, because it frankly seems buggy and does a poor job of that. I've thought of starting up Google Maps every now and then if the display is off (it would be annoying if I were actually using the phone at the time, so only when display is off) because that seems to immediately update Latitude. If I really wanted to get fancy I'd only do that when the battery was charged beyond some minimum threshold (which would be getting close to the algorithm that Google apparently failed to implement properly).
So lots of plans but only one simple working profile at the moment
