Is there a mechanism in Android, to know the direction in which the
physical keyboard would open on any device (left, right or up)?

No, not as far as I can see in the documentation (you did look through the official reference, right?).

That said, aren't all (current) slide-out keyboards attached to the portrait-left side of the screen? I mean, the stock OS only supports screen tilt in one direction (portrait->CCW->landscape->CW->portrait).

My observation is also the same that current android devices with portrait layout have slide-out keyboard attached to the left side of the screen.

I want to confirm whether android imposes some restriction on the direction in which slide-out keyboard will be attached to a android devices.
If no then can't we have android devices with portrait layout having slide-out keyboards attached to the right of the screen or android devices with landscape layout having slide-out keyboards attached to the bottom of the screen?
If we can have such devices, there should be some mechanism to determine the layout of the device and direction in which slide-out keyboard would open.

That's a bit of a chicken-or-egg situation, but if it's needed it will be in the OS. No need for you to worry about at this time. Android up to and including v2.1 can only tilt one way (and I suppose the same can be said for the iPhone).
